Young midfielder Will Hoskin-Elliott and former Swans forward Jed Lamb have been named in a 25-man squad to play Melbourne at Spotless Stadium this Sunday.
Zac Williams and Adam Tomlinson have also been added to the team which lost to St Kilda by seven points at Etihad Stadium last Saturday.
At this stage, Lachie Whitfield is the only omission with a foot injury.
21-year-old Lamb, who switched from the Swans during the off-season is yet to make his GIANTS debut, while Hoskin-Elliott, Williams and Tomlinson are also a chance to play their first AFL match for the season.
All four players impressed for the UWS GIANTS against the NT Thunder in the NEAFL last week.
Head Coach Leon Cameron said it was another exciting challenge for the GIANTS.
"The boys have trained well this week after a disappointing loss to St Kilda last weekend,” said Cameron.
"It’s a massive match this weekend up against the Melbourne Demons who are going to be hungry as they look for their first win of the season.
"The Demons were very competitive in round one against St Kilda and probably should have won that game, and we’re expecting a really close game on Sunday.
"It’s our last game in Sydney for six weeks so we hope to have a big crowd at Spotless Stadium to cheer on the GIANTS,” he said.
